Bonnot, Agnès, George Z. Mentis, Jesse Skoch, and Michael J. O’Donovan. Electroporation loading of calcium-sensitive dyes into the CNS. J Neurophysiol 93: 1793–1808, 2005. First published October 27, 2004; doi:10.1152/jn.00923.2004. Calcium imaging of neural network function has been limited by the extent of tissue labeled or the time taken for labeling. We now describe the use of electroporation—an established technique for transfecting cells with genes—to load neurons with calcium-sensitive dyes in the isolated spinal cord of the neonatal mouse in vitro. The dyes were injected subdurally, intravascularly, or into the central canal. This technique results in rapid and extensive labeling of neurons and their processes at all depths of the spinal cord, over a rostrocaudal extent determined by the position and size of the electrodes. Our results suggest that vascular distribution of the dye is involved in all three types of injections. Electroporation disrupts local reflex and network function only transiently ( 1 h), after which time they recover. We describe applications of the method to image activity of neuronal populations and individual neurons during antidromic, reflex, and locomotor-like behaviors. We show that these different motor behaviors are characterized by distinct patterns of activation among the labeled populations of cells.

A simple and fast method to image calcium activity of neurons from intact dorsal root ganglia using fluorescent chemical Ca2+ indicators

Chemical calcium indicators have been commonly used to monitor calcium (Ca2+) activity in cell bodies, i.e., somata, of isolated dorsal root ganglion neurons. Recent studies have shown that dorsal root ganglion somata play an essential role in soma-glia interactions and actively participate in the transmission of nociceptive signals. It is therefore desirable to develop methods to study Ca2+ ac...

Innovative Methodology Bulk electroporation and population calcium imaging in the adult mammalian retina

Briggman KL, Euler T. Bulk electroporation and population calcium imaging in the adult mammalian retina. J Neurophysiol 105: 2601–2609, 2011. First published February 23, 2011; doi:10.1152/jn.00722.2010.—The optical recording of light-evoked activity in populations of neurons in the mammalian retina offers several benefits over the use of multielectrode arrays. However, population imaging has b...

Loading a Calcium Dye into Frog Nerve Endings Through the Nerve Stump: Calcium Transient Registration in the Frog Neuromuscular Junction

One of the most feasible methods of measuring presynaptic calcium levels in presynaptic nerve terminals is optical recording. It is based on using calcium-sensitive fluorescent dyes that change their emission intensity or wavelength depending on the concentration of free calcium in the cell. There are several methods used to stain cells with calcium dyes. Most common are the processes of loadin...

Calcium Imaging of AM Dyes Following Prolonged Incubation in Acute Neuronal Tissue

Calcium-imaging is a sensitive method for monitoring calcium dynamics during neuronal activity. As intracellular calcium concentration is correlated to physiological and pathophysiological activity of neurons, calcium imaging with fluorescent indicators is one of the most commonly used techniques in neuroscience today. Current methodologies for loading calcium dyes into the tissue require prolo...
